Formulation

Thiamine mononitrate (Vit. B1), USP 300mg
Pyridoxine HCl (Vit. B6) 300mg
Cyanocobalamin (Vit. B12) 100mcg

Indications

For the treatment of B1, B6 and B12 deficiencies. Indicated for neuritis, neuralgia, shoulder-arm syndrome, facial paresis, pregnancy neuritis, drug-induced alcoholic and diabetic neuropathy and Vitamin B deficiencies.

Dosage and Mode/ Route of Administration

Prophylaxis: 1-2 tablets daily
Therapeutic: 2-4 tablets daily
to be taken orally

Contraindications

Hypersensitivity to the Vitamin B1, B6, and B12.

Warning

Administration of doses greater than 10mcg of Vitamin B12 daily may produce a hematological response in patients with folate deficiency, indiscriminate use may mask the precise diagnosis.

Pregnancy and Lactation

Supplementation did not significantly affect thiamine concentration in breast milk of healthy, well-nourished women when compared with those not given thiamine. Vitamin B6 and B12 are excreted and distributed into breastmilk respectively.

Interactions

Pyridoxine reduces the effects of levedopa but this does not occur if dopa decarbolase inhibitor is also given. Pyridoxine reduces the activity of altretamine. It has also been reported to decrease serum concentrations of phenobarbital and phenytoin. Many drugs may increase the requirements for pyridoxine: such as drugs include hydrazaline, isoniazid, penicillamine and oral contraceptives.

Absorption of vitamin B12 from the gastrointestinal tract may be reduced by neomycin, aminosalicylic acid, histamine H2- antagonists, omeprazole, and colchicine. Serum concentrations may be decreased by use of oral contraceptives.

Adverse Drug Reactions

Adverse effects of thiamine are rare.
Long-term use of large doses of pyridoxine is associated with the development of severe peripheral neuropathies.
